Effects of Epothilone A in Combination with Metformin and Sitagliptin in HepG2 Cells

چکیده
Studies indicate that diabetics are more likely to develop cancer, especially of the pancreas or colon, or hepatocellular carcinoma. Tumor genesis is promoted by high insulin and glucose levels pancreas, in T2DM (Saito et al., 2013; Zheng et al., 2013; Wang et al., 2015). Metformin, a commonly used drug for T2DM, reduces the incidence of various types of neoplasm. However, much less is known about the influence of the novel antidiabetic drug sitagliptin (SITA) on cancer occurrence, as well as its interaction with anticancer therapy. Additionally, the mechanisms of the possible interaction between antidiabetic drugs and anticancer agents have yet to be precisely determined. The aim of the present paper is to determine the influence of metformin (MET) and SITA on the killing of liver cancer cells mediated by epothilone A (EpoA), a microtubule inhibitor agent.

In-vitro Synergistic Effect of Metformin and Berberine on High Glucose-induced Lipogenesis
Metformin and berberine have been reported to have lipid lowering effects. This study aims to investigate lipid lowering effects of berberine and metformin, alone and in combination, in HepG2 cells to determine whether berberine and metformin work synergistically and elucidate their mechanisms. HepG2 cells were treated with 33 mM glucose in the presence of various concentrations of berberine an...

متن کاملSimultaneous Effects of Metformin and Sitagliptin on the Contents of Insulin Resistance Proteins Glucose Transporter 4 and Protein Kinase B in Diabetic Patients\' Adipose Tissue
Objective: Obesity is a factor in the development of insulin resistance and type 2 diabetes. Obesity contributes a wide variety of metabolic changes such as insulin resistance. The insulin signal mechanism to intra-cells occurs in insulin resistance, primarily in adipose tissue cells, which can be appropriate targets for therapeutic approaches by recognizing the proteins in this pathway. The st...
متن کاملBeneficial Endocrine but Adverse Exocrine Effects of Sitagliptin in the Human Islet Amyloid Polypeptide Transgenic Rat Model of Type 2 Diabetes
OBJECTIVE We sought to establish the extent and mechanisms by which sitagliptin and metformin singly and in combination modify islet disease progression in human islet amyloid polypeptide transgenic (HIP) rats, a model for type 2 diabetes. RESEARCH DESIGN AND METHODS HIP rats were treated with sitagliptin, metformin, sitagliptin plus metformin, or no drug as controls for 12 weeks. Fasting blo...
متن کاملEffects of quercetin on ionizing radiation-induced cellular responses in HepG2 cells
Background: Quercetin has been reported to modulate cell proliferation and apoptosis. The present study aimed at identifying whether treatment of ionizing radiation (IR) combined with quercetin induces apoptosis in HepG2 cells. Materials and Methods: HepG2 cells were plated at an appropriate density according to each experimental scale and irradiated with 1, 5 and 10 Gy gamma-rays from a 60Co s...
